Description

This antibody recognizes the CD5 antigen (T1 antigen) which is expressed on all mature T-cells, most thymocytes, and a subset of mature B-cells. Based on stimulatory effects of monoclonal antibodies, CD5 may be involved in signal transduction. Upon T-cell activation, CD5 is phosphorylated on tyrosine residues. CD5 crosslinking induces extracellular calcium mobilisation, tyrosine phosphorylation of intracellular proteins and DAG production. It may serve as a dual receptor, giving stimulatory or inhibitory signals depending both on the cell type and development stage. In thymocytes and B1a cells, CD5 leads to inhibitory signals, whereas in peripheral mature T-lymphocytes it acts as a costimulatory signal receptor. The antibody can be used for detecting CD5 in flow cytometry, frozen tissue sections, western blotting and immunoprecipitation. It also reacts with rhesus monkey lymphocytes.

Application Notes

FACS with whole blood: 100 μl blood + 10 μl antibody

FACS with cells: 500,000 cells in 50 μl + 10 μl antibody

This antibody recognizes the CD5 antigen (T1 antigen) on all mature T-cells, most thymocytes and on a subset of mature B-cells. In flow cytometry, the antibody stains 50-90% of human peripheral blood mononuclear cells.
