Cellular Antigens

Specificity of this antibody has been tested using Western blot, immunofluorescence and ELISA. The product reacts with human IgG (Fc) with no significant reactivity with human IgG (Fab2), IgM or other serum proteins. Other species IgG (Fc) were not tested.

CD45 antigen (Receptor-type tyrosine-protein phosphatase C, PTPRC) is a protein tyrosine-protein phosphatase required for T-cell activation through the antigen receptor. CD45 acts as a positive regulator of T-cell coactivation upon binding to DPP4. Defects in CD45 are a cause of severe combined immunodefiency autosomal recessive T-cell negative/B-cell-positive/NK-cell-positive (T-B+NK+ SCID). Genetic variations in CD45 are involved in multiple sclerosis susceptibility.

PEG10 (Paternally expressed gene 10 protein) participates apoptosis and cell differentiation pathways. PEG10 prevents apoptosis in hepatocellular carcinoma (HCC) cells through interaction with SIAH1, a mediator of apoptosis. PEG10 also plays a role in cell growth promotion and hepatoma formation. PEG10 inhibits the TGF-beta signaling by interacting with the TGF-beta receptor ALK1. When overexpressed, PEG10 induces the formation of cellular extension, such as filopodia in association with ALK1.

CD44 is a receptor for hyaluronic acid and can also interact with other ligands, such as osteopontin, collagens and matrix metalloproteinases (MMPs). CD44 function is controlled by its posttranslational modifications. One critical modification involves discrete sialofucosylations rendering the selectin-binding glycoform of CD44 called HCELL (for Hematopoietic Cell E-selectin/L-selectin Ligand). The HCELL glycoform was originally discovered on human hematopoietic stem cells and leukemic blasts, and was subsequently identified on cancer cells.

CD10 is a 100kDa glycoprotein, also designated Common Acute Lymphocytic Leukemia Antigen (CALLA). It is a cell surface enzyme with neutral metalloendopeptidase activity which inactivates a variety of biologically active peptides. CD10 is expressed on the cells of lymphoblastic, Burkitt’s, and follicular germinal center lymphomas, and on cells from patients with chronic myelocytic leukemia (CML). It is also expressed on the surface of normal early lymphoid progenitor cells, immature B cells within adult bone marrow and germinal center B cells within lymphoid tissue.
